On one’s toes は、「気を抜かない」「油断しない」という意味で使われているイディオムです。

On one’s toes を普通に訳すと、「つま先立ちで」ですね。足全体を地面に着けておくのではなく、つま先立ちでいることによって、必要があればすぐに行動に移せる、という意味合いです。

「即座に対応できるようにしておく」「いつでも動けるよう構えておく」と言いたいときに使える表現です。
